苗药理气活血滴丸对大鼠慢性心力衰竭的保护作用

Protective Effect of Miao Medicine Liqi Huoxue Dripping Pills on Chronic Heart Failure in Rats

目的:探讨苗药理气活血滴丸对异丙肾上腺素(ISO)诱导大鼠慢性心力衰竭(CHF)的保护作用。方法:将雄性SD大鼠随机分为2组:对照组和CHF模型组,利用皮下注射异丙肾上腺素的方法建立大鼠CHF模型。4周建模成功后,将大鼠随机分为模型组、理气活血滴丸低、中、高(43. 75、87. 50、175. 00 mg/kg/d)剂量组和依那普利(10mg/kg/d)组。药物干预4周后,采用酶联免疫法检测血清BNP水平,BL-420S生物机能实验系统监测大鼠左心室功能,TUNEL法分析心肌细胞凋亡情况。结果:与模型组比较,理气活血滴丸中、高剂量及依那普利组大鼠血清BNP水平、左心室舒张末压(LVEDP)和心肌细胞凋亡率显著降低,左心室收缩压(LVSP)、左心室内压最大变化速率(±dp/dtmax)显著升高。结论:理气活血滴丸对异丙肾上腺素致大鼠CHF的保护作用与其改善左心室功能及抑制心肌细胞凋亡有关。

Objective To abserve the protective effect of Miao medicine Liqi Huoxue dripping pills on isoproterenol( ISO) induced chronic heart failure( CHF) in rats. Methods The male SD rats were randomly divided into 2 groups: the control group and the CHF model group. The rat CHF model was established by subcutaneous injection of isoproterenol. After 4 weeks,rats were randomly divided into model group,Liqi Huoxue dropping pills including low,medium and high dose( 43. 75、87. 50、175. 00 mg/kg/d,respectively) groups and enalapril( 10 mg/kg/d) group. After 4 weeks of drug intervention,the serum BNP level was detected by enzyme-linked immunosorbent assay. The left ventricular function of rats was monitored by BL-420 S biological function test system.The apoptosis of myocardial cells was analyzed by TUNEL method. Results Compared with the model group,the serum BNP level,the left ventricular end diastolic pressure( LVEDP) and the apoptosis rate of myocardial cells were significantly decreased in the middle,high dose and enalapril group,and the left ventricular systolic pressure( LVSP) and the maximum change rate of left ventricular pressure( ± dp/dtmax) were significantly increased. Conclusion The protective effects of Liqi Huoxue dripping pills on isoprenaline induced CHF in rats are related to improving left ventricular function and inhibiting cardiomyocyte apoptosis.
