摘要
目的:建立白饭树中岩白菜素的高效液相色谱含量测定方法。方法:色谱柱为依利特Sino Chrom ODSBP (4. 6 mm×250 mm,5μm),流动相为甲醇-水(15∶85),流速1. 0 mL/min,柱温25℃,检测波长275 nm。采用正交设计法筛选供试品溶液的制备条件。结果:供试液制备条件为:药材破碎度为过四号筛,溶剂为100%甲醇,超声提取时间30 min。岩白菜素在0. 045 6~1. 519μg范围内有良好线性关系,平均回收率为102. 6%,RSD=2. 09%(n=6)。结论:本方法简便易行,结果准确可靠,可用于壮药白饭树中岩白菜素的含量测定方法。
Objective To establish a HPLC method for content determination of bergenin in Flueggae virosa Herba. Methods The HPLC with Elite Sino Chrom ODS-BP( 4. 6 mm × 250 mm,5 μm) was used;Methanol-water( 15∶ 85) was used as mobile phase at flow rate of 1. 0 mL/min;the detection wavelength was 275 nm and column temperature at 25 ℃. Orthogonal design was applied to study the preparation conditions of test sample solution. Result The optimal conditions were as followed: the samples were extracted by ultrasonic wave,the fragmentation degree was acrossing the fourth sieve,extraction solvent was 100% methanol,and extraction time was 30 min. The standard curve was linear in 0. 0456 ~ 1. 519 μg,the average recovery was 102. 6% with RSD of 2. 09%( n = 6).Conclusion The method is simple,accurate and reliable,which can be used to determine the content of of bergenin in Flueggea virosa Herba.
