摘要
目的:研究湘西山莓叶挥发油化学成分,及其抗氧化活性、抑菌作用,为山莓叶挥发油的进一步开发利用提供基础依据。方法:采用水蒸气蒸馏法提取山莓叶挥发油,通过气相色谱-质谱联用(GC-MS)技术确定其中化学成分与相对百分含量。运用1,1-二苯基-2-三硝基苯肼(DPPH)法评价山莓叶挥发油清除自由基能力,刃天青96孔板微量稀释法测试五种供试菌的抑菌作用。结果:鉴定山莓叶挥发油19种成分,占挥发油总质量的87.54%,其中酚类及含氧衍生物63.55%,烃类19.79%,酯类2.54%,醛类1.66%。山莓叶挥发油对DPPH自由基的清除呈量效关系,但效果弱于芦丁和Vc,对大肠杆菌、金黄色葡萄球菌、苏云金杆菌、沙门氏菌、痢疾志贺氏菌等五种供试菌均有抑菌作用,其中对痢疾志贺杆菌MIC为15.6μg/mL,效果明显好于卡拉霉素标准品。结论:山莓叶挥发油体外抗氧化能力相对较弱,对痢疾志贺杆菌抑制作用显著。
Object The chemical composition,antioxidant and antimicrobial activities of the volatile oil of Rubus corchorifolius L.f.leaves were investigated.Method The volatile oil was extracted by steam distillation,identified by GC/MS and determined by peak area normalization method.The antioxidant activity of the oil was tested by 1,1-diphenyl-2-picryl-hydrazyl(DPPH)method,the antibacterial activity was evaluated by renzurin microplate assay.Result A total of 19 components were identified,the main constituents were phenol derivatives(63.55%),alkanes(19.79%),esters(2.54%),aldehydes(1.66%)and etc.The volatile oil revealed a certain scavenging effect on DPPH free radical,the clearance effect was weaker than rutin and Vc.The volatile oil had a certain antibacterial effect on escherichia coli,staphylococcus aureus,bacillus thuring-iensis,salmonella,shigella dusenterlea.Minimum inhibitory concentration to shigella dusenterlea was 15.6μg/mL,that is lower than concentration of kalafungin standard.Conclusion volatile oil from Rubus corchorifolius L.f.leaves showed weak antioxidant and certain inhibition effect on shigella dusenterlea.
