摘要
目的 :了解PLGA经基因胜肽胶修饰后的促进骨髓间充质干细胞 (MSC)粘附作用。方法 :抽取成年兔骨髓 ,以密度梯度离心法获得间充质干细胞 ,置含 10 %小牛血清RPMI 164 0培养基培养。取第 3代MSC ,分别接种于 4块6孔培养板 ,其中 8孔预先放入包被PLGA膜的盖玻片作为对照组 ,8孔放入包被经基因胜肽胶处理的PLGA膜的盖玻片为实验组 ,剩余 8孔放入不做任何处理的盖玻片为空白对照组。分别于不同时间点检测细胞数 ,分析细胞的粘附情况。并采用免疫荧光技术观察细胞形态。结果 :结果显示基因胜肽胶能明显促进MSC在高分子材料上的粘附。 6h和8h时间点与对照组相比 ,有非常显著差异 (P <0 .0 1)。免疫荧光染色照片显示 ,实验组细胞以铺展型为主 ,对照组细胞明显皱缩。结论 :基因胜肽胶能促进MSC在高分子材料上的粘附。
Objective: The effect of Biotiegen,a novel adhesive protein,on the adhesion and morphology of rabbit bone marrow-derived mesenchymal stem cells(MSCs) was evaluated on PLGA.Methods:The third generation MSCs obtained from mature rabbits were cultured on PLGA coated by Biotiegen in RPMI1640 containing 10% fetal calf serum,while cultured on PLGA as control.The adhesive behavior of the cells at different times was analyzed by cell counting and morphology by FITC-conjugated immunofluorescence assay.Results:It was found that Biotiegen promoted cell adhesion on PLGA,which of the group coated by Biotiegen was significantly higher than that of control ( P <0.01) after 6 and 8h culture,meantime,the cells were better spreading after 6h culture compared with control.Conclusion:Biotiegen is effective in promoting the adhesion of MSC on PLGA.
