摘要
目的 :探讨头颈部恶性肿瘤组织中端粒酶活性水平及其与肿瘤发生、发展的关系。方法 :用改进的PCR TRAP法对 36例头颈部肿瘤组织、正常组织、癌旁组织分别进行端粒酶活性定量检测。结果 :36例头颈部恶性肿瘤中有 32例 (88.9%)阳性表达 ,癌旁组织有 4例 (11.1%)阳性表达 ,正常组织有 1例 (2 .8%)阳性表达 ,肿瘤组织与癌旁组织和正常组织之间 ,差异有显著性意义 (P <0 .0 5 )。端粒酶活性水平与临床分期、病理类型和分化程度不呈线性相关 ,癌旁组织和正常组织阳性病例的端粒酶定量水平较肿瘤组织低。 结论 :端粒酶的激活与头颈部恶性肿瘤的发生、发展可能密切相关 ;端粒酶活性有可能成为恶性肿瘤分子诊断的标志物和治疗的新靶点。
Objective:To determinate quantitatively the level of telomerase activity in human head and neck malignant tumors and to analyze their relationship with clinical factors.Method:Telomerase active level in 36 cases of malignant tumors , adjacent and normal tissues was determined by modified PCR-TRAP assay.Result:The positive rate of telomerase in tumor tissue, adjacent and normal tissues was 88.89%, 13.89%, 2.78%,respectively. we found :Although there is different TPG units in 32 positive cases, the level of telomerase activities is independent of the following factors: clinical stages, differentia pathological types and degrees. Simultaneously, we also found that there was a higher telomerase level in tumor than that in normal and adjacent tissues (P< 0.05).Conclusion:These data show that there is a high expression of telomerase in head and neck malignant tumors. The activated telomerase may be related to the generation and progression of carcinoma, and theoretically may be as a new biological marker and therapeutic target for these tumors.
