摘要
目的 :观察妊娠高血压综合征 (妊高征 )患者胎盘组织中蛋白质的差异表达 ,筛选出与该病相关的特异性表达的蛋白质分子 ,从而为阐明该病发病机制奠定基础。方法 :应用双向电泳技术 ,对 10例正常妊娠妇女及10例妊高征患者胎盘组织总蛋白质进行分离 ,得到蛋白质图谱。利用相关软件进行图像分析 ,筛选出妊高征患者差异表达的蛋白质 ,并利用质谱指纹对差异蛋白加以分析。结果 :提取胎盘组织总蛋白 ,运用双向凝胶电泳方法展示了妊高征患者胎盘中的差异蛋白 ,电泳图谱显示共有 5 0 5种蛋白质 ,其中 ,有 16种蛋白从质和量上在妊高征患者胎盘组织中表达增高。结论 :妊高征患者胎盘组织中存在一些差异蛋白 ,这些蛋白质分子与磷脂酶C有关 ,PLC介导的信号传导途径在妊高征的发生过程中可能有重要作用 ;双向电泳是适合筛选差异蛋白的技术。
Objective:To screen the differentially expressed proteins in placenta tissue of patients with pregnancy induced hypertension (PIH). Methods:Two-dimensional polyacrylamide gel electrophoresis was used to display the total proteins of placenta of 10 patients with PIH and 10 healthy pregnancy women. We analysed the two-dimensional gel images by relative software and looked for the differentially expressed proteins compared with normal placenta. Results: We displayed and screened the differential proteins in the placenta tissue of PIH patients.Conclusions:The differential proteins are relative to Phospholipase C -γ 2 (PLC-γ 2) .The results suggest that the signal transmission mediated by PLC be a role in the mechanism of PIH; two -dimensional gel electrophoresis is a suitable method to screen different proteins.
