摘要
目的 :为探讨ras基因突变及P2 1蛋白表达与膀胱癌发生发展关系。方法 :采用聚合酶链反应 -单链DNA构象多态性分析和免疫组化链霉菌抗生物素蛋白 -过氧化物酶法 ,对 30例膀胱癌组织Ha -ras基因ExonⅠ突变及P2 1蛋白表达情况进行了检测。结果 :膀胱癌组织Ha -ras基因ExonⅠ突变率为 4 6 .7% ,高于其周边组织的 16 .7% ,经 χ2 检验 ,差异有显著性 (P <0 .0 2 5 ) ,其 p2 1蛋白表达率前者为 6 0 .0 % ,也明显高于其周边组织 2 6 .7% (P <0 .0 1)。结论 :Ha -rasExonⅠ点突变及其P2 1蛋白的过度表达在膀胱癌的发生发展中起着重要作用。
Objective:To investigate the relationship between the bladder cancer and ras gene mutation or P 21 oncoprotein overexpression.Methods:Thirty cases of bladder cancer specimens were analysed by polymerase chain reaction product-single strand conformation polymorphism analysis and streptavidin peroxidase conjugated method.Results:Ha-ras Exon Ⅰ mutation and p 21 protein expression ratio: 46.7% and 60.0% in bladder cancer were higher than 16.7% and 26.7% in normal tissue around the cancer tissue ( P <0.025 and P <0.01 respectively ).Conclusion:Ha-ras Exon Ⅰ mutation and p 21 protein overexpression play an important role in the origin and development of the bladder cancer.
