摘要
目的 :探讨细胞因子IL 1α和IL 10与凋亡相关基因Bcl 2家族中Bax和Bcl 2在 1 甲基 4 苯基 1,2 ,3,6 四氢吡啶(MPTP)致C5 7BL6J小鼠帕金森病模型中黑质和纹状体内的表达意义及关系。方法 :利用RT PCR的方法检测对照组 (NS组 )C5 7BL6J小鼠及皮下注射MPTP后第 6天C5 7BL6J小鼠的左右侧脑内黑质、纹状体、下丘脑内的IL 1α ,IL 10 ,Bax和Bcl 2的mRNA的表达。结果 :与对照组相比 ,IL 1αmRNA在帕金森小鼠模型纹状体内表达显著增加 ;除对照组 1只外 ,对照组和MPTP组均未检测到IL 10mRNA的表达 ;BaxmRNA和Bcl 2mRNA在黑质内的表达均显著低于对照组。结论 :黑质和纹状体在MPTP所致的小鼠帕金森病中受累 ,表现为有不同基因的消长。促进炎症发生的细胞因子IL
Objective:To study the mRNA expression of cytokines(IL 1α and IL 10)and apoptotic genes(Bax and Bcl 2) in C57BL6J mice after the administration of 1 methyl 4 phenyl 1,2,3,6 tetrahydropyridine(MPTP).Methods:C57BL6J male mice received a single subcutaneous injection of MPTP(40mg/kg).Control group was injected with NS.On the sixth day after injection,the mRNA expression of IL 1α,IL 10,Bax and Bcl 2 in the substantia nigra,striatum and hypothalamus was studied by RT PCR.Results:As compared to the NS group,IL 1α mRNA expression was significantly higher in the striatum of mice injected with MPTP,but not in the substantia nigra and hypothalamus.IL 10 mRNA expression was not detected in any brain structures from both groups.As compared to the NS group,Bax mRNA and Bcl 2 mRNA expression in the substantia nigra,but not in the striatum and hypothalamus,were significantly lower in the MPTP group.Conclusion:The mechanisms of MPTP induced Parkinson disease include both inflammation and apoptosis.The expression of the cytokine(IL 1α) increase in the striatum,but its role is not elucidated yet.Mitochondria pathway of apoptosis in the substantia nigra may be involved in the MPTP induced Parkinson disease.
