摘要
目的 :运用基因芯片技术研究骨肉瘤转移相关基因表达 .方法 :提取具有不同转移特性的骨肉瘤细胞总RNA ,反转录制备杂交探针 ,应用基因芯片筛选与骨肉瘤转移相关的基因 .杂交信号用ScanArray 30 0 0扫描 ,结果用ImaGene3.0软件分析 .结果 :对原位及转移骨肉瘤细胞基因表达谱分析 ,发现两种肿瘤细胞中存在差异表达基因 ,其中有抑癌基因、转移相关蛋白基因、细胞周期蛋白基因等 ,与肿瘤发生发展密切相关 .结论
AIM: To study the expression of osteosarcoma metastasis associated gene with cDNA microarray. METHODS: Total RNA of low metastatic osteosarcoma (SOSP 9607) and high metastatic osteosarcoma (SOSP M) was extracted, purified to mRNA and then reverse transcripted to cDNA respectively. The cDNA of SOSP 9607 was labeled with Cy3 and the cDNA of SOSP M was labeled with Cy5. The two samples were hybridized with the cDNA microarray. The hybridization signals were scanned by ScanArray3000 and analyzed by software ImaGene 3.0. RESULTS: Differentially expressed genes were found between the two different metastatic potential cells, such as tumor suppression genes, metastasis associated protein gene and cell cycle protein gene. They were all correlated to tumor occurrence and development. CONCLUSION: This study provides a new approach to identifying genes that may be associated with metastasis.
