摘要
目的 研究粉尘螨浸液的变应原成分及其活性。方法 取实验室人工培养的粉尘螨 ,制备粉尘螨浸液 ,以SephadexG -75、SephadexG -2 0 0凝胶柱层析 ,收集各管洗脱液进行皮肤挑刺试验。 结果 粉尘螨浸液经SephadexG -75柱层析后 ,洗脱液的A值呈现双峰 ,能引起皮肤红晕或风团的洗脱液主要位于两峰之间接近第 1峰处 ;经SephadexG -2 0 0柱层析后呈 3峰 ,能引起皮肤红晕或风团的洗脱液主要位于两峰之间接近第 2峰处。结论 具有免疫学活性的粉尘螨变应原位于第 1峰、第 2峰及两峰之间 ,是免疫诊断和治疗所需组分。
Objective To study on antigenic fractions and immunocompetence of Dermatophagoides farine extract.Methods After chromatography on Sephadex G-75 and Sephadex G-200, elution was used in skin prick test (SPT).Results After Dermatophagoides farine extract passed through gel filtration, it was found that the optical densities (A) of the fractions had 2 peaks on Sephadex G-75 and SPT-positive elution was near to the first peak between the two peaks. However, the A of the fractions had 3 peaks on Sephadex G-200 and SPT-positive elution was near to the second peak between the first and second peaks.Conclusion Immunocompetence of Dermatophagoides farine extract is located at the first peak, the second peak and between them, which are all needed to be used in immunotherapy and diagnosis of allergic diseases.
