摘要
目的 探讨洛伐他汀对阿霉素肾病大鼠肾小球硬化的作用及机制。方法 16只Wistar大鼠在阿霉素注射制备肾病模型后高脂饮食 ,并随机分为高脂组及治疗组 ,治疗组予洛伐他汀 4mg/kg ,饲养 12周 ,观察血脂、尿蛋白、肾脏病理改变。结果 治疗组血胆固醇、甘油三酯、低密度脂蛋白、尿蛋白、肾脏病理积分及泡沫细胞明显低于高脂组 ,肾脏免疫组化显示治疗组转化生长因子 β(TGF β1)、纤维连接蛋白 (FN)减少。结论 洛伐他汀可减轻高脂血症造成的肾小球损伤 ,其机制之一为减少TGF β1在肾脏中的表达。
Objective To determine the effect and mechanism of lovastatin on glomerulosclerosis in adriamycin induced nephri tic rats.Methods Sixteen male Wistar nephritic rats induced by adriamycin were randomly divided into two groups, the treatment group and hyperlipidemia group. They were fed 12 weeks. Urinary protein excretion and serum lipid were assayed, and renal glomerulosclerosis index, the expression of transforming growth factor β 1 (TGF β 1) and fibronection(FN) were observed.Results Serum total cholesterol, trigtycerides ,low density lipoprotein were significantly lower in treatment group than that in hyperlipidemia group from 4 weeks. At 12 weeks , the renal glomerulosclerosis index , the expression of TGF β 1 and FN were also sharply lower in treatment group than that in hyperlipidemia group.Conclusion Our finding indicated that lovastatin can lesion glomerular injury that result from hyperlipidemia , and one of its mechanism is that decreasing the expression of TGF β 1 in renal tissue.
