细胞周期抑制蛋白p27^(kip1)和周期蛋白表达与大肠癌发生预后(英文) 被引量:13

摘要 目的探讨细胞周期调控抑制因子p27kip1蛋白和周期蛋白E(cyclinE)在大肠腺癌发生发展中的作用。方法采用免疫组化技术SP法,检测p27kip1和cyclinE蛋白在37例大肠腺癌和26例正常大肠黏膜组织中的表达。结果p27kip1蛋白阳性表达率在大肠腺癌组织中为45.9%,显著低于正常大肠黏膜组织(χ2=4.590,P<0.05),并与大肠腺癌组织分化程度和淋巴结转移均有关(χ2=9.653,χ2=6.838,P<0.05);cyclinE蛋白阳性表达率在大肠腺癌组织中为64.9%,显著高于正常大肠黏膜组织(χ2=4.285,P<0.05),并与大肠腺癌组织分化程度和淋巴结转移均有关(χ2=8.397,χ2=5.261,P<0.05);p27kip1蛋白阳性表达组cyclinE蛋白阳性表达率低于p27kip1蛋白阴性表达组,但两者无相关(r=-0.1133,P>0.05)。结论p27kip1蛋白的低表达和/或cyclinE蛋白的高表达在大肠腺癌发生中发挥重要作用,并可能有助于预测其恶性程度及淋巴结转移,可能对大肠腺癌的治疗和生活质量的改善有指导作用。 Aim The relationships between p27 kip1 protein as negative factor of cell cycle regulation and c olorectal adenocarcinoma have been sel-domly reported in China.This article aims to investigate the effects of i n-hibiting factor of cell cycle regula tion-p27 kip1 and cyclinE proteins in the genesis and progression of human colorectal adenocarcinoma.Methods The expressions of p27kip1and cyclinE p roteins in tumor tissues of 37patien ts with colorectal adenocarcinoma and in normal colorectal mucosal tissues of26subjects were detected by SP immunohistochemical technique.Results p27 kip1 protein positive-expression rate i n tumor tissues of colorectal adeno-carcinoma was 45.9%,which was lower than that in normal colorectal mucosal tissues(χ 2 =4.590,P<0.05),p27 kip1 protein positive-expression correlated significantly with tumo r cell differentiation and lymph nod e metastasis(χ 2 =9.653;χ 2 =6.838,P<0.05);cyclinE positive-expression rate in tumor tissues was 64.9%,which was higher than that in normal colorectal mucosal tissues(χ 2 =4.285,P<0.05);cyclinE posi-tive-expression also correlated significantly with tumor cell differe ntiation and lymph node metastasis(χ 2 =8.397;χ 2 =5.261,P<0.05);cyclinE pro-tein positive-expression rate in th e tumor tissues of p27 kip1 protein posi-tive-expression group was lower than that in p27 kip1 protein nega-tive-expression group,there were n o significantly correlation betwee n the two groups(r=-0.1133,P>0.05).Conclusion The decreased expres-sion of p27 kip1 protein and /or over -expression of c yclinE protein may playan important role in the genesis of colorectal adenocarcinoma and might b e helpful to the determination of mali gnant degree and lymph node metastasis as well as the treatment and living quality of the diseases.
