摘要 目的为了正确使用氯化钙于可注射组织工程骨之中,研究不同浓度氯化钙对骨髓基质成骨细胞增殖与分化的影响。方法从兔髂骨中获取骨髓基质成骨细胞,将骨髓基质成骨细胞置于含不同浓度氯化钙的细胞培养液中,培养24,72,120h后,通过检测骨髓基质成骨细胞的代谢活性和碱性磷酸酶活性。观察不同浓度氯化钙对成骨细胞增殖和分化的影响。结果骨髓基质成骨细胞在不同浓度(1~10mmol)的氯化钙培养液中培养24,48,72h后,除1mmol浓度氯化钙在细胞培养72h后对成骨细胞增殖稍有促进作用外,其余各组随着细胞培养液中氯化钙浓度的升高,骨髓基质成骨细胞的代谢活性逐渐降低。骨髓基质成骨细胞在不同浓度(1~10mmol)的氯化钙培养液中培养24,72,120h后,随着氯化钙浓度的增加,成骨细胞中碱性磷酸酶活性逐渐降低,各组与对照组间差异均有显著性意义(P<0.01)。结论氯化钙对骨髓基质成骨细胞增殖与分化有一定的抑制作用。 Aim For correct using calcium chloride i n injectable tis-sue-engineered bone,we study effec t of calcium chloride on proliferati on and differentiation of osteoblastic ce lls.Methods The osteoblasts were isolated from iliac bone of New Zealand rabbits,then were cultured in DMEM medium with different concentrations of calcium chloride.After 24,72,120h cu lture,effect of different levels of calciu m chloride on proliferation and diff erentiation of osteoblastic cells were assessed by the measurement of cellular metab olism activity and alkaline phosphate act ivity.Results After the osteoblasts were cultured in DMEM medium with differe nt concentrations of calcium chloride(1-10mmol)for 24,72,120h,with the concentrat ion of calcium chloride rises in the medium,cellular metabolism a ctivity of every experimental groups de-cline except 1mmol calcium chloride for 72h,which had a little promoting effect.With the concentration of ca lcium chloride rises in the medium,alkaline phosphate activity decline.The difference between experimental group and control group was statistically sig nificant (P<0.01).Conclusion Calcium chloride can inhibit proliferation and differentiation of osteoblastic cells.
