摘要 目的 探讨牙周炎患者唾液多胺水平的变化,建立正相高效液相色谱的测定方法。方法 牙周炎患者唾液经纯化、丹磺酰氯衍生化后,用苯提取,以Hypersil BDS CN为正相色谱柱,丙酮-氯仿(2:98,V/V)为流动相,检测波长为425 nm,流速1 ml/min。 结果 多胺中腐胺和精胺在50~1 000 nmol/ml范围内线性关系均良好(r=0.999 1;r=0.999 2),本法回收率分别为94.65%和90.48%,RSD为1.6%和2.1%;牙周炎患者唾液中多胺水平明显增高。结论 本法灵敏、稳定,重现性好,检测病人唾液中多胺含量具有临床实用意义。 Objective To develop a high-performance liquid chromato-graphic method for the determination of polyamine in patients' saliva with periodontitis. Methods The derivatization with dansyl chloride was followed by normal - phase chromatography with a mobile phase of acetone /chloroform (2:98,v/v) at a flow rate of 1ml/min and the detection wavelength was set at 405nm. Results The standard curve was linear over the concentration range of 50-1000nmol/ml for putrescine and spermine with correlation coefficient 0.9991 and 0.9992. The recoveries were 94.65 % with RSD 1.6% and 90.48 % with RSD 2.1% for putrescine and spermine, respectively. The putrescine and spermine content in patients' saliva with periodontitis was much higher than that in normal humans ( P < 0.01) . Conclusion The present study provided a sensitive, accurate and reproducible method for the determination of polyamine levels in human saliva.
