乳果糖对严重烫伤大鼠血浆循环内毒素及组织中白细胞介素-18mRNA基因表达的影响

摘要 目的探讨乳果糖对烫伤大鼠组织中白细胞介素-18 mRNA表达的影响。方法将大鼠致30%体表面积Ⅲ度烫伤,随机分成烫伤对照组(C组)及乳果糖预防组(L组)。采用逆转录PCR技术检测肠、肺、肝、肾等组织中IL-18 mRNA水平。结果烫伤后两组体循环内毒素含量显著升高,用乳果糖予处理后可显著降低循环内毒素含量(P<0.05或P<0.01)。烫伤后,肠、肺、肝、肾等组织中IL-18 mRNA表达较伤前显著升高(P<0.01),伤后8 h达峰值,24 h内居高不下。给予乳果糖予处理后可明显抑制IL-18 mRNA的过度表达(P<0.05或P<0.01)。结论乳果糖可明显降低体循环内毒素的含量,并明显抑制肠、肺、肝、肾组织中IL-18 mRNA的过度表达。 Objective To investigate the effect of oral lactulose on interleukin-18 mRNA expression.Methods Rats were subjected to a full-thickness thermal injury of 30% body surface area.The injured rats were randomly divided into two groups:scald controls (group C) and lactulose treatment group (group L) .IL-18 mRNA expressions in tissues of intestine, lung,liver and kidney were determined by reverse transcription polymerase chain reaction (RT-PCR). Results The endotoxin contents in systemic circulation was significantly elevated in two groups.There were much lower LPS contents in the group L ( P <0.05) .IL-18 mRNA expressions in the intestine ,liver and kidney rapidly increased after injury ( P < 0.01),peaking at 8 hours and maintained a high level until 24 h. However,after treatment with lactulose, the LPS contents were lowered and IL-18 mRNA expressions were markedly inhibited in various organs. Conclusions Lactulose decreases the IPS contents in systemic circulation and markedly inhibits excessive IL-18 mRNA expression in vital organs.
