摘要 目的:研究黄芪多糖(APS)在长期骨髓细胞培养中是否能刺激或支持体外造血作用及刺激基质细胞的增殖.方法:采用长期骨髓细胞培养法,研究APS对骨髓细胞的影响.结果:在APS作用下,培养体系中的非贴壁细胞数在5~8周增加19~168倍.第6周时,在APS处理的培养体系中,97% 非贴壁细胞为成熟中性粒细胞,而对照仅为8%.实验结束即第17周时,APS使贴壁细胞数从每瓶2.2×105增加到4.1×105.结论:黄芪多糖在长期培养骨髓细胞(LTBMC)中可支持体外造血作用. Objective : To investigate whether Astragalus Polysaccharide (APS) can stimulate or maintain hematopoiesis in vitro in long term bone marrow culture (LTBMC), and whether APS can stimulate the proliferation of bone marrow stromal cells. Method : LTBMC was established using the method of Dexter in the study. Result: APS significantly increased the number of non-adherent cells in the cultures by 19-168 fold from week 5 to week 8. On week 6, 97% of the non-adherent cells from the APS treated cultures were mature neutrophil, while the control cultures had only 8% neutrophils. APS treated cultures had a much higher number of adherent stromal cell layer (4.1 × 105) than that of the control cultures (2.2 × 105) by the end of the LTBMC (week 17). Conclusion: APS can support hematopoiesis in vitro.
