摘要
目的 研究Gq 1 1 α蛋白在氯丙烯引起的周围神经病中所起的作用。方法 原代培养的乳大鼠大脑细胞染毒 2 4h ,用WesternBlot方法和免疫组化技术分析测定Gq 1 1 α蛋白含量的变化。结果 免疫组化结果显示在乳大鼠大脑细胞中 ,氯丙烯使Gq 1 1 α蛋白含量显著升高 ,低剂量组升高 19% ,高剂量组升高 3 4% (P <0 0 1) ;WesternBlot结果显示氯丙烯使Gq 1 1 α蛋白含量显著升高 ,低剂量组升高 2 4% ,高剂量组升高 3 9% (P <0 0 1)。结论 提示氯丙烯急性中毒可导致原代培养的乳大鼠大脑细胞膜的Gq 1 1 α蛋白相对含量升高 ,上调Gq
Objective In order to acquire the mechanism of allyl chloride (AC)induced toxic peripheral neuropathy in human.Methods The G q/11 α was analyzed with Western Blotting and immunohistochemistry technology after the primary cultured cells of cerebrum in new born rats were treated with AC for 24 hours.Results By using immunohistochemistry method,G q/11 α was increased 19% in 0\^6 mmol/L group and 34% in 1\^2 mmol/L group respectively( P <0\^01) compared with controls.And it was increased 24% in 0\^6 mmol/L group and 39% in 1\^2 mmol/L group ( P <0\^01) by Western Blotting.Conclusion The results suggest that AC can increase the contents of G q/11 α of primary cultured cells in rat's cerebrum,and regulate up G protein\|PLC signal transduction pathway.
