摘要
目的 探讨 p2 7KIP1与原发性肝细胞癌 (HCC)发生、发展的关系。方法 应用SABC(链霉素亲和素 生物素复合物 )免疫组化染色和mRNA原位杂交方法 ,对 5 2例肝细胞肝癌中p2 7KIP1蛋白和mRNA的表达进行了检测。结果 p2 7KIP1基因的表达主要位于肝或肝癌细胞的细胞核中 ,呈细颗粒状分布。p2 7KIP1蛋白在癌旁肝组织中的阳性细胞指数为 ( 10 .7± 7.6) % ,明显高于癌组织的 ( 1.5± 0 .9) % ,并且 p2 7KIP1蛋白表达的阳性细胞指数与HCC细胞的分化程度呈正相关。p2 7KIP1mRNA杂交阳性物呈蓝色细颗粒状 ,主要位于肝或肝癌细胞的细胞核和细胞浆中 ,mRNA阳性的细胞分布更广泛 ,但在癌旁肝组织和不同分化程度的HCC组织中 ,p2 7KIP1mRNA的阳性细胞指数差异无显著性意义。结论 p2 7KIP1蛋白与HCC的发生及细胞分级有关。
Objective To study the relationship between expression of p27 KIP1 and progression of hepatocellular carcinoma(HCC).Methods The expression of p27 KIP1 in 52 cases of HCC was detected by immunohistochemistry of strept avidin biotin complex and mRNA in situ hybridization.Results The positive cells of p27 KIP1 protein were diffused in HCC.The positive signal was localized in nuclei.The labeling index (LI) of p27 KIP1 protein was significantly higher in tumor surrounding tissues than that in tumor tissues. p27 KIP1 protein LI showed a positive correlation with the differentiation grade of HCC.The better differentiation of cancer cells, the higher LI of p27 KIP1 protein ( P <0.01).The positive cells of p27 KIP1 mRNA were also diffused in HCC.The positive signal was localized in nuclei and cytoplasm. As to the expression of p27 KIP1 at the mRNA level,there was no significant correlation with tumor surrounding tissues and stages of HCC.Conclusionp27 KIP1 protein is associated with progression and differentiation grade of hepatocellular carcinoma.
