以玉米芯和麸皮 ( 7∶3 )为碳源 ,(NH4) 2 SO4( 2 % )为氮源 ,3 0℃培养 72h ,发酵曲用蒸馏水3 0℃浸提 1h ,得到 β 1,4 木聚糖酶活力高 ,β 木糖苷酶活力低的粗酶液。β 1,4 木聚糖酶和 β 木糖苷酶的最适作用温度分别为 5 0℃和 60℃ ,最适作用pH分别为 4 8和 4 0 ,β 1,4 木聚糖酶在pH5 0~ 10 6范围内稳定 ,β 木糖苷酶在 pH 3 0~ 3 0范围内稳定。β 木糖苷酶的热稳定性比 β 1,4
To obtain xylanase preparation with high level endo xylanase and low level β xylosidase, the conditions of soli state fermentation of the Aspergillus niger M 1 were optimized as follows: corncob and wheat bran (mass fraction 7∶3) as carbon source, 2%(NH 4) 2SO 4 as nitrogen source, cultivated at 30℃ for 72. After cultivation the media were extracted at 30℃ for 1h with distilled water. Study on enzyme properties showed that the optimal reactio pH and temperature of xylanase and β xylosidase were pH4.8, 50℃ and pH4.0, 60℃, respectively. The xylanase and β xylosidase were stable in the pH range 5.0~10.6 and 3.0~7.0, respectively. The thermostabilities of two enzymes were also studied showing that the β xylosidase was more thermostable than the xylanase.
