摘要
目的 研究NGF对丙烯酰胺引起小脑神经元细胞凋亡的影响。方法 建立丙烯酰胺诱导的小脑神经细胞凋亡模型 ,并利用腹腔注射方法观察NGF的保护作用。结果 从行为学检测丙烯酰胺诱导神经细胞凋亡后自主行为活动情况 ,结果显示模型对照组自主性行为的活动次数明显减少。生化指标模型对照组与实验组比较 ,模型对照组大鼠的小脑组织中谷光甘肽和肌酸激酶含量明显减少 (P <0 0 1 )。从形态学观察模型对照组小脑皮质蒲肯野细胞层和颗粒细胞层凋亡明显 ,细胞数明显减少。NGF (1 0 0 0 ,2 0 0 0u kg)能够使谷光甘肽和肌酸激酶含量增加 ,细胞形态结构损伤明显减轻。结论 NGF能够抑制丙烯酰胺诱导的小脑神经细胞凋亡。
Objective To investigate the effect of NFG on cerebellar neuronal apoptosis induced by acrylamide. Methods The model of cerebellar neurnal apoptosis was induced by acrylamide in rats.The protective effect of NGF was observed. Results The frequency of voluntary movements in control group was much lower than that of NGF-treated group,( P <0 01).Compared with NGF-treated group,the neuronal apoptosis in control group was obvious.The patterned loss of cerebellar,Purkinnje cells and cerebellar granule cells in control group were much higher than that of NGF-treated group. Conclusions The data indicate that NGF could reduce recebellar neuronal apoptosis induced by acrylamide.
