慢性乙肝患者树突状细胞表型和抗原提呈能力的测定

DETERMINATION OF PHENOTYPE AND ANTIGEN PRESENT ABILITY OF DC IN PATIENTS WITH CHRONIC HEPATITIS B

目的 :探讨慢性HBV感染者DC的表现和功能及其与免疫耐受的关系。方法 :选择正常人和慢性HBV感染者 ,从外周血中分离单个核细胞 ,用细胞贴壁法富集DC ,流式细胞术测定DCHLAⅡ和共刺激分子的表达 ,同位素掺入法测定DC的抗原提呈能力 ,ELISA方法测定抗原提呈过程中Th1和Th2类细胞因子的水平。结果 :慢性HBV感染者HLAⅡ和共刺激分子表达水平、抗原提呈能力和Th1类细胞因子的水平均低于正常 (P <0 .0 5 ) ,而Th2类细胞因子的水平与正常相似 (P >0 .0 5 )。结论 :慢性HBV感染免疫耐受与DC的缺陷有关。

Objective:To understand the relationship between dendritic cells (DC) and immunotolerance in chronic persistent hepatitis B virus (HBV) infection.Methods:Thirteen normals and 23 patients were included in the study.DCs were enriched from peripheral blood mononuclear cells by density grade centrifugation and panning.HLA Ⅱ and costimulatory molecules on the surface of DCs were measured by flow cytometry.The antigen presenting activity of DCs was determined by 3 H-TdR incorporation.During this process,the productions of cytokines were evaluated by enzyme linked immunosorbent assay.Results:The express levels of HLA Ⅱ and costimulatory molecules on DCs derived from the patients were lower than those from the normal( P <0.05),as well as the antigen presenting activity and the Th1 class cytokines( P ,0.05).Conclusion:The pathogenesis of immunotolerance in chronic persistent HBV infection is associated with the defect of DCs.
