摘要
目的 :探讨糖尿病新西兰兔血清肿瘤坏死因子 (TNF -α)和一氧化氮 (NO)在糖尿病中的变化以及它们在糖尿病发病机制中的作用。方法 :采用高脂高糖饲料喂养新西兰兔 ,建立一种新的糖尿病动物模型。观察糖尿病新西兰兔血清中的一氧化氮水平 ,一氧化氮合酶 (NOS)活力及血清肿瘤坏死因子水平的变化。结果 :糖尿病新西兰兔出现大量蛋白尿 (P <0 .0 5 ) ;肌酐清除率 (CCr)明显增高 (P <0 .0 0 1) ;血清中肿瘤坏死因子、NO水平和NOS活力明显增高 (P <0 .0 0 1) ;血糖及胰岛素明显增高 (P <0 .0 0 1)。结论 :糖尿病新西兰兔血清肿瘤坏死因子、NO水平和NOS活力明显增高 ,糖尿病的起因可能与它们的异常变化有关。
Objective:To study the change of tumor necrosis factor-α and nitric oxide(NO) in the serum of diabetic New Zealand White rabbits and the role of them in diabetes. Methods:Two groups of New Zealand White rabbits received regular rabbit chow (the normal control) or high fat high sucrose diet for 4 months. The levels of plasma insulin and glucose were investigated.Level of NO and NO synthase (NOS)activity and level of tumor necrosis factor-α in serum were measured.Results: In test group it existed higher tumor necrosis factor-a, NO level and NOS activity than control group( P <0.001).There was an elevation of the kidney/body weight in the early stage of the diabetes( P <0.05)as well as a great mount of proteinuria( P <0.05).The creatinine clearance rate(CCr)increased apparently( P <0.001). Conclusion: Level of tumor necrosis factor-α, NO and NO synthase activity in serum of New Zealand white rabbits increased.The cause of diabetes may be related with these abnormal changes.
