支气管哮喘病人CD8^+T细胞CD25,CD30表达状况

THE EXPRESSION BOTH CD25 AND CD30 ON CD8A+T LYMPHOCYTES IN ASTHMATIC PATIENTS

目的 :通过观察哮喘病人外周血CD8+ T细胞CD2 5 ,CD30表达水平 ,了解哮喘病人CD8+ T细胞功能状态。方法 :将所分离出的CD8+ T细胞分别用PPD ,PHA刺激 ,最后用流式细胞仪检测细胞表面CD2 5 ,CD30表达水平。结果 :PHA刺激组CD8+ T细胞CD2 5表达水平高于健康对照 (P <0 .0 5 ) ,而CD30表达则无差异 ;PPD刺激组CD8+ T细胞CD2 5 ,CD30表达与健康对照组相比无显著差异 ;哮喘病人自然状态下 (即未受抗原刺激 )CD8+ T细胞CD2 5表达与健康对照无差异 ,而CD30表达低于健康对照 (P <0 .0 5 )。结论 :哮喘病人CD8+ T细胞活化状态明显异常。

Objective:To evaluate the expression levels both CD25 and CD30 on CD8 + cells in asthmatic patients.Methods:T cells isolated from peripheral blood mononuclear cells were separated by nylon column measurement. CD8 +T cells were gotten by negation selection using magnetic beads; and then, they were stimulated by PPD and PHA; finally, CD25 and CD30 on CD8 +T cells were detected by flow cytometor. Results:In PHA-stimulated group, CD25 levels on CD8 +T cells were as higher than that of healthy controls; but CD30 expression was not different;The expression both CD25 and CD30 was no statistic significance compared with healthy controls in PPD-stimulated group.The levels both CD25 and CD30 without antigen stimulation in asthmatic patients were significantly lower than that of healthy controls ( P <0.01 or P <0.05 ). Conclusion:The activated state of CD8+T cells are abnormal in asthmatic patients.
