枯草杆菌溶血纤维蛋白酶发酵培养基配方的确定 被引量:3

ASCERTAINING THE MEDIUM INGREDIENTS FOR A FIBRINOLYTIC ENZYME SECRETING BACILLUS SUBTILIS STRAIN

摘要 采取有交互作用的正交试验设计了枯草杆菌HL986菌株溶血纤维蛋白酶发酵的培养基成分 ,用试管表面培养法和摇瓶培养法进行发酵 ,利用牛血纤维蛋白琼脂平皿法测溶血纤维蛋白酶的活性 ,分析确定的最佳培养基配方为 :进口酵母浸出粉OXOID2 % ,麦芽糖 0 .2 % ,NaH2 PO4·2H2 O 0 .7%。 Orthogonal tests were used to ascertaining the factors and levels of the optimum medium for a fibrinolytic enzyme producing Bacillus subtilis natto strain(HL986).Sequential surface cultivation and shaking incubation were used.Fibrinolytic activity of the enzyme was examined by disk diffusion test on bovine fibrin,and metered by urokinase activity unit.The results indicated that,the optimum medium ingredient was 2% Oxiod yeast extract powder,used as non-protein nitrogen source;0.2% maltose;and 0.7%NaH 2PO 4·2H 2O.
