都柏林念珠菌的鉴定及其方法学研究 被引量:1

摘要 〔目的〕都柏林念珠菌和白色念珠菌的生物学特性非常相似 ,两者常常混淆 ,研究其常规的生化特性及运用特异性基因扩增的方法 ,探索其生物学及分子生物学间的差异 ,寻找一种简便的鉴别方法。〔方法〕用常规方法分离并鉴定都柏林念珠菌和白色念珠菌 ,如 45℃生长试验 ,木糖利用试验等 ,以白色念珠菌细胞色素P45 0L1A1基因设计一对特异性引物 ,做PCR基因扩增试验。〔结果〕初步分离的 3 96株疑似白色念珠菌 ,经生化鉴定发现有 9株生物学特性符合都柏林念珠菌 ,做PCR基因扩增试验 ,3 87株实验菌株均发现有 2 43bpDNA条带 ,而 9株都柏林念珠菌则无。〔结论〕常规生化试验和PCR基因扩增试验方法用于都柏林念珠菌和白色念珠菌的鉴别诊断简便可靠。 睴bjective〕 Candida dubliniensis and Candida albicans are usually confounded with each other for their biology characters are extraordinarily similar. There fore, i t is necessary to search of the biologic and molecular biologic differences of them and establish a simple and veIiable method to distinguish Gandida dubliniensis from Candida albicans by using routine biochemistry character or specific gene amplification. 〔Methods〕 Use routine methods, for example, Growth at 45℃ for 48h and Xylose utilization, to separate and identify Candida dubliniensis from Candida albicans. Use the polymerase chain reaction (PCR) with a couple of primers from the cytochrome P450L 1A 1 of Candida adbicans. 〔Results〕 In suspected 396 Candida albicans, 9 strains were coincidence with Candida dubliniensis by using biochemical identification. With PCR amplification, 387 Candida albicnns laboratorial isolates can produce the 243bp DNA product, but 9 Candida dubliniensis can’t.〔Conclusion〕 It’s a simple and reliable method of using the routine bio-chemical test and PCR in identifying the Candida dubliniensis from the Candida aibicans.
