摘要
分离纯化乌骨藤提取物成分,通过建立荷人宫颈腺癌裸鼠模型,研究其抗肿瘤生物活性作用。采用硅胶色谱方法从乌骨藤提取物中分离纯化得到两个皂苷晶体,利用光谱方法进行了结构确定。通过裸鼠灌胃方法检测乌骨藤皂苷晶体Ⅰ、Ⅱ对荷人宫颈腺癌裸鼠肿瘤抑制作用。乌骨藤晶体Ⅰ、Ⅱ分别被鉴定为11α-O-顺芷酰-12β-O-乙酰通关藤苷元B-3-β-O-D-葡萄吡喃糖基-(1→4)-6-去氧-3-O-甲基-D-阿洛吡喃糖基-(1→4)-D-夹竹桃吡喃糖苷(Ⅰ)、11α-O-顺芷酰-12β-O-乙酰通关藤苷元B-3-β-O-D-葡萄吡喃糖基-(1→4)-β-O-D-葡萄吡喃糖基-(1→4)-6-去氧-3-O-甲基-D-阿洛吡喃糖基-(1→4)-D-夹竹桃吡喃糖苷(Ⅱ)。通过荷人宫颈腺癌裸鼠的肿瘤抑制实验,显示乌骨藤皂苷Ⅰ、Ⅱ对肿瘤生长具有明显抑制作用(P<0.01)。皂苷类成分具有抑制肿瘤生长功效,该类成分在乌骨藤植物中含量高,具有重要的开发研究价值。
The study aimed to separat and purify the saponin form Marsdenia tenacissima ,and researching further investigateon the antitumor biological activities through establishment of transplanted human cervi-cal cancer in nude mice model.The compounds were isolated by silica gel chromatography,which were then identified by the spectroscopy.The compounds Ⅰ,Ⅱ were detected with inhibitory effects on human cervi-cal cancer in nude mice through gavage methods.Two compounds were identified as 11-O-tigloxyl-12β-O-ethanoyl tenacigenin B 3-O-β-D-glucopyranosyl-(1→4)-6-deoxy-3-O-methyl-β-D-allopyranosyl-(1→4)-β-D-oleandropyranoside (Ⅰ),11α-O-tigloxyl-12β-O-ethanoyltenacigenin B-3-β-O-D-glucopyranosyl-(1→4)-β-O-D-glucopyranosyl-(1→4)-6-deoxy-3-O-methyl-β-D-allopyranosyl-(1 →4)-β-D-oleandropyranoside(II).They present obvious inhibitory effects after inhibitory experiment through human cervical cancer in nude mice. CONCLUSION:Saponins possessed to inhibit tumor growth,they were high content and important devel-opment value.
