摘要 改进和优化保健品及特殊膳食食品中维生素B_6的前处理方法及液相色谱条件。样品经10g·L^(-1)偏磷酸溶液提取等前处理后,C18柱分离,荧光检测器检测,外标法定量分析维生素B6(吡哆醇、吡哆醛和吡哆胺)的含量;色谱条件:色谱柱:C18反相色谱柱(粒径5μm,150mm×4.6mm);流动相:乙腈50mL,庚烷磺酸钠2.0g,三乙胺2.5mL,用水溶解并定容到1000mL(冰乙酸调pH至3.1);流速1.0mL·min-1;柱温30℃;检测波长:激发波长293nm,发射波长395nm。维生素B6采用本文建立的方法,维生素B6的3个组分出峰时间缩短为10min以内,线性范围是0.01~2.00μg·mL-1,最低检出限2.5μg/100g。方法的回收率为93.1%~105.6%,RSD小于5.0%。此方法适用于保健品及特殊膳食中维生素B6的检测,同时简化了前处理过程,缩短了检测时间,节省了检测试剂,对环境污染减轻,且分离效果很好。 To improve and optimize the pretreatment method and HPLC conditions to determine vitamin B6 in healthcare products and special diet food.Food samples were extracted in 10 g·L-1 of Metaphosphoric acid solution,separated with C18 columns and then detected with fluorescence detector.The samples were finally analyzed with external standard method for content of vitamin B6 (pyridoxine,pyridoxal and pyri-doxamine).Chromatographic conditions:C18 column (5 μm,150 mm× 4.60 mm)was used as stationary phase.The mobile phase was composed with 50mL acetonitrile,2.0 g Heptane sulfonic acid sodium,2.5 mL triethylamine and capacity to 1 000 mL with water (pH 3.1 by Ice acetic acid);The flow rate was 1.0 mL ·min-1 with the column temperature at was 30℃.Excitation wavelength was 293 nm,emission wave-length was 395 nm.Results Method of vitamin B6 in this paper with a proportion the appearance time in 3 groups were shortened to 10 min.Excellent linearity of vitamin B6 appeared at a concentration of 0.01 -2.00 μg·mL-1 .The detection limit for pyridoxine,pyridoxal and pyridoxamine all was 2.5 μg/100 g.The recovery rate was 93.1%-105.6%,with RSD<5.0%.This method could be suitable for determination all kinds of vitamin B6 in foods and At the same time simplify the pretreatment process,saving detection rea-gent,alleviation of environmental pollution and good separating effect.
