摘要
目的 建立新的HIV亚型分析方法 ,并检测其特异性及敏感性。方法 HIV 111个亚型及HIV 2的env基因C2 V3区一段核苷酸序列 ,用芯片点样机点样于经预处理的玻片 ,制备成HIV亚型分析基因芯片 ,并用其对 15份HIV阳性样本进行亚型分析 ,所得结果与目前常用HIV亚型分析方法 基因序列分析法所得结果进行比较。结果 制备的基因芯片对 15份HIV阳性样本检测结果为 :样本 0 1、0 2、0 3、0 5、0 8、0 9、11、12、13、15为HIV 1B亚型 ;0 6、0 7为HIV 1C亚型 ;0 4、10、14为HIV 1E亚型。结论 制备的基因芯片亚型分析结果与基因序列分析法所得结果完全符合 ,敏感性高、特异性好 ,并且具有操作简单、省时、价廉等特点 ,有望推广应用。
Objective To set up a new method-genechip to determine the subtype of HIV-1 and putted it into practical use to investigate its value of diagnosis.Methods The character sequences of HIV-env gene C2-V3 section were enlarged and dotted on chips and 15 HIV positive samples were analysized by these genechips.The results were contrasted with standard HIV subtype deternination methods-gene sequences analysis.Results Sample No.01,02,03,05,08,09,11,12,13,15 were confirmed as HIV-1 subtype B;No.06,07 were confirmed as HIV-1 subtype C;No.04,10,14 were confirmed as HIV-1 subtype E.The results were consistent with the results got by standard HIV subtype deternination method-gene sequences analysis.Conclusion This genechip was sensitive and specific for HIV subtype determination,it can be used widely as a new,efficient and cheap HIV-1 subtype detecting method for clinic diagnosis.
