银屑病患者外周血单个核细胞Th_1/Th_2细胞因子表达研究

Expression of Th_1/Th_2 Cytokines in the Peripheral Blood Mononuclear Cells of the Patients with Psoriasis

目的 探讨Th1/Th2 细胞因子在银屑病发病机理中的作用。方法 应用逆转录 聚合酶链反应法 (RT PCR)半定量分析 19例正常对照和 2 2例银屑病患者外周血单个核细胞中TNFα、IFN γ和IL 4、IL 10mRNA的表达。结果 银屑病患者TNFα和IFN γ表达显著上调 (P <0 .0 1) ,而IL 4显著降低 (P <0 .0 1) ,IL 10较正常降低但无显著差异(P >0 .0 5 )。结论 Th1型细胞因子在银屑病患者中过度表达 ,可能提示银屑病免疫病理机理中的T淋巴细胞活化模式。

Objective To investigate the pathogenic effects of Th 1/Th 2 cytokines in psoriasis.Methods Using RT-PCR technique to semiquantitatively analyse the expression of the mRNA expression of TNFα,TFN-γ and IL-4,IL-10 in the PBMCs from 19 normal controls and 22 psoriatic patients.Results The mRNA expression of TNFα and IFN-γ in psoriasis was upregulated compared with normal controls(P<0.001),but the expression of IL-4 mRNA was significantly decreased.There was no significant difference,though IL-10 was decresed compared with nomal controls(P>0.05).Conclusion The overexpression of Th 1-type cytokines within the psoriatic group may indicate the mode of T lymphocyte activation contributing the immunopathogenesis of psoriasis.
