Determination of isoproturon in water by enzyme-linked immunosorbent assay

摘要 建立了测定水中异丙隆的酶联免疫吸附法 .灵敏度 0 97μg L ,最低检出限 0 0 3μg L .16种取代脲类除草剂的交叉反应率低于 2 2 % .水样可不经分离提取直接测定 .10 %甲醇、2 %乙腈和 2 %乙醇不影响测定 .比较了 11种与异丙隆结构相近的半抗原的活性 . A direct enzyme-linked immunosorbent assay (ELISA) for the determination of isoproturon in water was developed. The concentration of isoproturon that inhibits 50% of antibody-antigen binding (IC 50 ) was 0.97?μg/L. The determination limit (IC 90 ) was 0.03?μg/L. Sixteen urea herbicides showed negligible cross-reactivity (<2.2%). The cross-reactivities of the 16 haptens showed that the selected antiserum had high activity. Up to 10% of methanol, 2% of ethanol, and 2% of acetonitrile could be used in the assay. The assay has been successfully applied to the determination of isoproturon in water samples.
