超声波-甲醇法提取茶皂素工艺研究 被引量:9

摘要 以茶粕为原料,采用超声波-甲醇法从茶粕中提取茶皂素.分别考察了甲醇浓度、料液比、提取温度、提取时间等因素对茶皂素的提取率和纯度的影响.单因素试验结果表明:甲醇浓度90%、料液比1∶5、提取温度30℃和提取时间30 min时为宜.通过L9(34)正交试验进一步考察发现:甲醇浓度对茶皂素的提取率和纯度影响较大,提取时间和料液比分别对提取率和纯度影响较小.从实际应用考虑,选取甲醇浓度90%,提取温度30℃,料液比1∶7及提取时间30 min为适宜的工艺条件,此时,茶皂素提取率和纯度分别为96.21%和59.3%,该研究为茶粕的应用提供了一种新的可行方法. Taking tea seed meal as the raw material,the technology that tea saponin is extracted from tea seed meal by ultrasonic assisted methanol extraction was studied in the paper.Based on the extraction efficiency and the purity of tea saponin from tea seed meal,effects of methanol content,mass ratio of solid to liquid,extraction temperature and extraction time were systematically investigated.The results obtained from the s ingle-factor experiments show that the suitable condition was methanol content of 90%,solid-to-liquid ratio of 1∶5,extraction temperature of 30°C and extraction time of 30 min.With the othorgofial experiment of L9(34),the influencing factors of the extraction of tea saponin were further inspected,and the results indicate that methanol concentration has great influence on the extraction efficiency and purity of tea saponin,while the ex traction time and mass ratio of solid to liquid has an inferior effect on the extraction efficiency and purity of tea saponin.In consideration of the practical application,the condition,such as methanol concentration of 90%,extraction temperature of 30°C,mass ratio of solid to liquid of 1∶7 and extraction time of 30 min,was used to extract and purify the tea saponin from the tea seed meal,under which the extraction efficiency and the purity of the tea saponin were 96.21%and 59.3%,respectively.Therefore,a feasible method referred in the paper was established to utilize the tea seed meal.
