摘要
设计了小麦印度腥黑穗病菌 (Tilletiaindica)和黑麦草腥黑穗病菌 (Tilletiawalkeri)通用引物H4 /H7和H11/H12 ,结合T .indica特异性引物Tin3/Tin4和T .walkeri特异性引物Tin11/Tin4建立了检测T .indica和T .walkeri冬孢子的套式PCR (nestedPCR)检测方法。检测的灵敏度达 1个冬孢子 ,检测时间缩短为 8h。PCR产物的测序验证了PCR反应的可靠性。这一方法克服了冬孢子休眠或不具存活力以及常规PCR检测时间长的困难 。
A simple and practical nested PCR method using primers H4/H7, H11/H12,Tin3/Tin4 and Tin11/Tin4 was developed for detecting teliospore of Tilletia indica and Tilletia walkeri .One teliospore can be amplified and obtained specific band.The process of detection can be finished within 8 hours.Because of avoidance of DNA extraction,the method can be used for detection of dormant teliospore of T.indica and T.walkeri.
