窖泥中产酸功能菌筛选及发酵条件初探 被引量:4

摘要 从洋河酒厂优质老窖泥中分离纯化筛选出一株产己酸的芽孢杆菌YHY03,经生理生化分析初步鉴定为克氏梭状芽孢杆菌。通过正交优化获得最优发酵培养基,并对YHY03发酵产酸代谢过程进行研究。实验结果表明:其发酵改良培养基的物质最优配比为:1.5%蔗糖;2%乙醇;0.5%乙酸钠,0.1%酵母膏、0.04%磷酸氢二钾、0.05%硫酸铵、0.02%硫酸镁。优化后己酸产量为285.32mg/100m L,己酸产量提高比例为148.2%,优化效果较明显。该菌株代谢产生己酸主要通过两个途径:0至4天直接利用乙酸合成,同时积累丁酸;6至12天利用丁酸反应形成己酸。 A caproic acid bacteria strain named YHY03 was isolated from century- old pit mud in Yanghe Distiller. Such strain was classified as Clostridium Kluyverii in biological properties through preliminary identification. The best fermentation medium was obtained by orthogonal experiment. And the fermentation process of YHY03 was studied to analyze the pathways of producing acid metabolic. The results show that: such strain was classified as Clostridium Kluyverii in biological properties through preliminary identification. The best medium for fermentation was composed of 1.5% sucrose 2% ethanol; 0.5% sodium acetate;0.1% yeast extract;0.04% potassium hydrogen phosphate; 0.05% ammonium sulfate; 0.02% magnesium sulfate.The optimization effect is obvious that the caproic acid production was285.32 mg/100 m L, increased 148.2%. The synthesis of caproic acid mainly through two ways.From 0 to 4 days, the acetic acid was directly consumed to synthesis of caproic acid and accumulation of butyric acid. From 6 to 12 days, the caproic acid was synthesized with butyric acid.
