摘要
目的:研究牛磺胆酸(Taurocholic Acid,TCA)对瞬转TGR5受体的HEK-293T及NR8383细胞的c AMP含量的影响。方法:采用MTT法测定TCA对细胞毒性范围,采用ELISA法测定TCA对细胞内c AMP含量的影响。结果:TCA对瞬转TGR5受体的HEK-293T细胞产生的c AMP含量呈剂量依赖性增加(P<0.05),TCA对瞬转TGR5受体的NR8383细胞产生的c AMP含量显著增加(P<0.05)。结论:TCA对TGR5介导的两种细胞的c AMP的含量均有显著性提高。
It is important to study effects of Taurocholic acids( TCA,a kind of natural bioactive substance of animal bile acids) on c AMP production in HEK- 293 T cells or NR8383 cells expressed TGR5 for its comprehensive pharmacological effects. Cell toxicity of TCA was measured by MTT; Intracellular c AMP production in cells expressed TGR5 was measured by ELISA. TCA increased the production of c AMP in cells induced by TGR5( P < 0. 05),and TCA dose- dependentlyin HEK- 293 T cells,but not in NR8383 cells. The potency of Taurine- conjugated lithocholic acid( TLCA) was higher than TCA on increased c AMP in HEK293 T cells( P <0. 05). TCA increased the production of c AMP in cells induced by TGR5 associated with cell types.
