欧李仁抗氧化肽的免疫功能及其表征结构的研究 被引量:5

STUDY ON THE IMMUNE REGULATION FUNCTION AND STRUCTURAL CHARACTERIZATION OF THE Cerasus Humilis Kernel ANTIOXIDANT PEPTIDE

摘要 本文研究了经三酶(碱性蛋白酶、胰蛋白酶、中性蛋白酶)复合水解制备的欧李仁抗氧化肽的免疫调节作用以及确定欧李仁抗氧化肽的表征结构。通过给正常小鼠灌胃不同剂量的欧李仁抗氧化肽来研究其对正常小鼠的免疫调节作用;并利用葡聚糖凝胶Sephadex G-25层析法对制备的欧李仁肽进行分离纯化并收集各纯化组分,将对DPPH·自由基的清除能力最强的组分进行表征结构的鉴定。试验表明欧李仁抗氧化肽能提高小鼠免疫器官指数,还可以增强小鼠淋巴细胞的增殖能力以及因绵羊红细胞致敏而出现的肿胀反应,还可促进小鼠血清溶血素和溶血空斑的形成以及小鼠的碳廓清能力。欧李仁多肽液经葡聚糖凝胶Sephadex G-25分离纯化后主要获得有4个组分,且在相同浓度下(2mg/ml)第4个组分对DPPH·自由基的清除能力最强,其清除率可达95. 6%,经质谱鉴定这个组分有2个肽片段分别为Leu-Ala-Thr-Ala-Pro-Ser-Arg和Leu-Asp-Gln-Val-Pro-Arg。研究表明欧李仁抗氧化肽对正常小鼠具有较好的免疫调节作用。 Immunoregulation function of the Cerasus Humilis Kernel peptide prepared by enzymatic hydrolysis and the structural characterization were inverstigated. The immunoregulation function was determined depend on the effect of the Cerasus Humilis Kernel peptide on the development of immune organs,cellular immunity,humoral immunity and mononuclear macrophage function of the mice;Purified the Cerasus Humilis Kernep peptide using Sephadex G-25 chromatography and collected all the components. Identificated the primary structure of the group which have the strongest scavenging ability of DPPH free radical. The Cerasus Humilis Kernel peptide could improve the index of thymus and spleen,enhance the transformation of lymphocytes,and swelling reaction of mice sensitized by sheep erythrocytes. In addition,the Cerasus Humilis Kernel peptide could promote formation of serum hemolysin and hemolytic plaque,and also could increase the ability of carbon clearance of the mice;four main components of Cerasus Humilis Kernel peptides were separted and obtained by Sephadex G-25 chromatography. The DPPH free radical scavenging capacity of the fourth components was the strongest at the same concentration( 2 mg/ml),its scavenging rate can reach 95. 6%,this group consists of two peptide fragments by the mass spectrometric identification. They were Leu-Ala-Thr-Ala-Pro-Ser-Arg and Leu-Asp-Gln-Val-Pro-Arg. These findings suggest that the Cerasus Humilis Kernel antioxidant peptide has favorable effects on enhancing immune function.
