摘要
目的 :用反相高效液相色谱 (RP HPLC)法测定环孢素脂质体乳胶剂中环孢素A的含量。方法 :以SpherisorbC8( 5 μm ,2 0 0mm× 4.6mmID)为色谱柱 ,乙腈 pH 2磷酸盐缓冲液( 68∶3 2 )为流动相 ,检测波长UV 2 10nm ,流速 1.0ml·min-1,柱温 60℃。结果 :环孢素A在 10~ 40 0mg·L-1浓度范围内线性关系良好 ,回收率为 99.5 2 % ,相对标准差为 1.62 % (n =5 )。结论 :RP HPLC法准确 ,灵敏度高 ,重现性好 。
Objective A rapid and sensitive reversed phase high performance liquid chromatographic method for the determination of cycloporine A in cycloporine A liposomes cream has been developed.Methods The mixture of pH2 potassium phosphate buffer?acetonitrile(68∶32) was used as a mobile phase,while a Spherisorb C 8 column (5?μm,200?mm× 4.6 ?mm?ID) was used as a stationary phase, and CsA was detected at 210?nm with flowing rate being rate 1.0ml·min -1 and column temperature at 60?℃.Results CsA had a good linear relation in the range of 10~400?mg·L -1 ,the mean recovery of was 99.52 %,the RSD was 1.62%( n =5).Conclusion The result showed that this method was reliable and accurate and it can be applied to the content determination of cycloporine A in cycloporine A?liposomes cream.
