摘要
目的 :研究人工合成葡糖基转移酶GTF第 5 5 2~ 5 70位的多肽疫苗HDS的免疫原性。方法 :将HDS与大分子载体钥孔帽贝血蓝蛋白偶联后 ,在腮腺区皮下免疫大鼠 ,取鼠血清 ,Westernblot法检测鼠血清抗GTF抗体。结果 :硝酸纤维膜上 4 3KD 6 6KD间出现强阳性条带 ,此条带位置与葡糖基转移酶经SDS PAGE电泳后条带位置一致。结论 :抗人工抗原HDS KLH抗体能与葡糖基转移酶发生抗原抗体反应 ,抗HDS
Objective:To investigate the antigenicity of the peptide vaccine HDS(552~570) from Streptococcus mutans glucosyltransferase(GTF).Methods:HDS was conjugated with keyhole limpet hemocyanin,a kind of carrier protein,to form a large molecule protein HDS KLH,which was injected in the vicinity of rats.The sera of rats was collected and Western blot assay was done to demonstrate whether there were specific antibodies to recognize GTF in rats anti HDS KLH serum.Results:There appeared an positive strip between 43kd~66kd in the fibrous membrane,which related to findings in SDS PAGE.Conclusion:The ability of anti HDS KLH recognizing GTF was demonstrated in this study.
