摘要
目的:观察温阳活血退黄方对阴黄证大鼠肝细胞凋亡,相关调控基因bcl-2、bax蛋白表达的影响。方法:72只大鼠随机分为6组:正常对照组,阴黄证模型组,阳黄对照组,阴黄证模型加温阳活血退黄方高、低剂量组,茵陈术附汤组。采用TUNEL和免疫组化法检测大鼠肝细胞凋亡和bcl-2、bax蛋白表达。结果:阴黄模型组大鼠肝细胞凋亡程度明显高于阳黄模型组及正常对照组(P<0.01),温阳活血退黄方高剂量组肝细胞凋亡程度明显低于阴黄模型组(P<0.05)。温阳活血退黄方高、低剂量组肝组织bcl-2蛋白表达明显高于阴黄模型组(P<0.01,<0.05),且其bax蛋白表达明显低于阴黄模型组(P<0.01,<0.05),体现较好量效关系。结论:温阳活血退黄方可能通过促进bcl-Z蛋白表达抑制bax蛋白表达阻断阴黄证大鼠肝细胞凋亡,可能是其治疗阴黄证的机制之一。
Objective: To investigate the effect of Wenyang Huoxue Tuihuang Electuary on hepa-tocyte apoptosis and the expression of its apoptosis-regulating gene bcl-2 > bax in rat model with Yin jaundice. Methods: Seventy-two rats were divided randomly into 6 groups: the normal control group, Yin jaundice model group, Yang jaundice model group, high-dosage treatment group, low-dosage treatment group, control treatment group. TUNEL assay and immunohistochemistry technique were used to detect the apoptosis of hepatocytes and the expression of bcl-2, bax in hepatocytes respectively. Results: The rate of apoptosis cells in Yin jaundice model group was significantly higher than that in Yang jaundice model group and normal control group ( P <0. 01) , and the rate of apoptosis cells in high-dosage treatment group was significantly lower than that in Yin jaundice model group ( P <0. 05). The expression of bcl-2 in high-dosage and low-dosage treatment group and control treatment group was significantly higher than that in Yin jaundice model group ( P <0. 01 or P <0. 05), and the expression of bax in these groups was significantly lower than that in Yin jaundice model group ( F <0. 01 or P <0. 05). Conclusion: Wenyang Huoxue Tuihuang electuary could prevent hepatocyte apoptosis perhaps by upregulating the expression of bcl-2 and downregulating the expression of Bax, which might be one of its mechanisms in treating Yin jaundice.
