通腑法对大鼠肠源性肺损伤保护作用机制的研究 被引量:24

Expermental Study on the Mechanism of Protective Effect of Clearing the Hollow Fu-organs on the Lung's Damage Caused by Partly Ligating the Large Intestine

摘要 目的:探讨肿瘤坏死因子(TNF-α)、内毒素(ET)在肠源性肺损伤发病过程中的作用及通腑利肺的作用机制。方法:将40只SD大鼠随机等分为正常对照组、模型组、解扎组及治疗组,每组各10只,采用体外直肠不全结扎法造模。采用动态浊度法检测各组血浆ET水平,放射免疫分析法检测各组血清TNF-α含量,RT-PCR法检测肺组织TNF-α mRNA水平的变化,并将通腑治疗组、解扎组与模型组及正常对照组对照。结果:模型组血浆ET及血清TNF-α水平明显高于正常对照组(P<0.01),肺组织TNF-α mRNA表达明显增高(P<0.01);通腑法治疗后ET水平、TNF-α含量和TNF-α mRNA表达均低于模型组(P<0.01),且肺损伤程度减轻。结论:ET、TNF-α在肠源性肺损伤过程中起重要作用,通腑法对损伤肺组织起保护作用。 Objective: To explore the effect of TNF-α on the lung's damage camsed by ET from partly ligated large intestine, and the influence of clearing the hollow fu-organs on the expression of TNF-α mRNA. Methods:Forty SD rats were radomly divided into four groups:Normal group,model group, ligation group and treatment group( n =10 each) ,the model was made by the method of partly ligating the recta outside the body. The level of LPS in blood plasma was measured by dynamic neph-elo metric method and the content of TNF-a in blood serum was detected by the method of radioim-munity. The expression of TNF-a was detected by RT-PCR method. And the results of each group were be compared. Results:The level of blood plasma ET,blood serums TNF-a of the model group and the expression of TNF-a mRNA in the lung tissue were remarkably higher than those of the normal group ( P <0.01). After the treatment of clearing the hollow fu-organs, all of the indexes decreased and the results were lower than those of the model group,and the degree of the lung's damage decreased. Conclusion: TNF-α plays a very important role during the lung's damage caused by ET from partly ligated large intestine, and clearing the hollow fu-organs can protect the lung from being damaged.
