摘要
我们用高效液相色谱(HPLC)研究了抗生素类抗癌药物丝裂霉素C(MMC)的酸解反应动力学.MMC在酸性介质中水解,生成三种产物.该酸解反应属一级平行反应.本文给出了介质pH值、浓度和温度对酸解速率常数的影响规律.对高效液相色谱分离出来的酸解产物作了红外光谱和吸收光谱研究.还讨论了MMC酸解反应的机理.
Mitomycin C (MMO) is an antideoplastic antibiotic drug. In this paper, in order to explore its pharmocology processes of anticancer effect in human body, its kinetics of reaction processes and separation of reactive products in acidic solution have been investigated by high performance liquid chromatography (HPLC). Hydrolysis of MMC in an acidic solution produces three products and the reaction belongs to a first-order reaction. Temperature, pH and concentration of solution not only have influence on the ratio of products of acidolysis, but also on the constant of reaction rate in the process of acidolysis. Using Fourier IR analysis, a study on acidolysis products separated by HPLC indicated that MMC eleminated its methoxy at first and then opened its aziriding ring and made a adding reaction to add a hydroxyl or an acid radical.
