摘要
目的 研究重组人血小板型磷脂酶 A2 (PL A2 )在杀菌过程中与常用抗菌药间相互作用关系 ,为临床用药提供依据 ,并探讨其杀菌机制。方法 将重组人血小板型 PL A2 和抗菌药分别单独以及联合作用于金黄色葡萄球菌 ,通过测定细菌菌落数来评价杀菌效果。结果 青霉素、头孢拉定、磷霉素、万古霉素增强血小板型 PL A2 杀菌活性 ;红霉素减弱血小板型 PL A2 杀菌活性 ;氧氟沙星对血小板型 PL A2 杀菌作用无影响。结论 血小板型 PL A2与破坏细胞壁的杀菌类药物有协同作用 ,与抑菌类药物合用受到拮抗 ;细菌细胞壁的结构及其生长状态是影响血小板型 PL A2
OBJECTIVE To study the interactions of recombinant human platelet phospholipase A 2 (PLA 2) and antibacterial drugs in their bactericidal process, for providing evidence for clinical application and exploring bactericidal mechanism of the PLA 2. METHODS Staphylococcus aureus was treated by PLA 2 and antibacterial alone or in both combination. Antibacterial activity was determined by counting colony forming units of bacteria. RESULTS Benzylpenicillin, cefradine, fosfomycin and vancomycin significantly increased bactericidal activity of recombinant human platelet PLA 2. Erythromycin inhibited the bactericidal activity of PLA 2. Ofloxacin had no effect on PLA 2 bactericidal activity. CONCLUSIONS The effects of the PLA 2 bactericidal activity were synergistic with cell wall active antibacterial drugs, antagonistic with bacteriostatic antibacterial drugs; the cell wall structure of bacteria and the state of cell growth are important factors in bactericidal activity of the PLA 2.
