老年癫痫大鼠海马NMDA受体亚单位1mRNA表达的研究 被引量:2

摘要 目的 研究老年癫痫大鼠海马 NMDA受体亚单位 1 (NMDAR1 ) m RNA表达的变化 ,为进一步探明癫痫发病及损伤的分子生物学机制奠定基础。方法 采用在立体定位仪将海人酸注射至老年大鼠杏仁核制备癫痫模型。将老年大鼠随机分为正常对照组 ,假手术组及致癫痫组 ,分别于不同时间取材 ,进行脑组织 NMDAR1 m RNA原位杂交检测。结果 正常海马各区均有极少量的 NMDAR1 m RNA阳性细胞分布 ;致癫痫组NMDAR1 m RNA水平显著高于正常对照组及假手术组 (P<0 .0 1 )。结论 大鼠海马各区都存在着 NMDAR1 m RNA不同程度的表达 ,癫痫后老年大鼠海马 NMDAR1 m RNA的表达水平上调。 Objective To observe the expression of NMDAR1 in hippocampus of the aged epilepsy rats.Methods the aged epilepsy rat models were made by injecting kainic acid into amygdala under stereotactic instrument,60 male Wistar rats were divided randomly into 6 groups. 1 the normal control group;2 the false operation group;3-6 epilepsy group.NMDAR1mRNA in situs hybridization of brian tissue were tested .Results In the normal group and false operation group,NMDAR1 positive cells distributed rarely in hippocampus;NMDAR1mRNA positive cells of epilepsy groups were significantly higher in hippocampus CA1,CA3,and dentate gyrus.Conclusions NMDAR1mRNA expressed in rat hippocampus in different degree,they were up-regulated in the aged epilepsy rats .
