昆虫病原线虫和共生细菌培养系统中噬菌体的检测 被引量:2

DETECTION OF LYSOGENIC PHAGE IN THE CULTURE OF SYMBIOTICXENORHABDUS AND PHOTORHABDUS BACTERIA AND SOLIDCULTURE SYSTEM OF ENTOMOPATHOGENIC NEMATODES

摘要 本文利用紫外照射、温度、pH、盐度诱导五株溶源性共生菌株Xenorhabdus和,同时测定斯氏和异小杆属线虫Steinernema carpocapsae A24,S.carpocapsae ALL,S.feltiae English,S.feltiae SN,Heterorhabditis bacterophora H06在体外培养过程中是否存在感染共生细菌的噬菌体。结果未发现噬菌斑,说明实验菌株在实验诱导条件下以及昆虫病原线虫固体培养系统中不可能诱发噬菌体的危害。 The effect of temperature (at 45C for a period from 0.5 to 4.5hours, and at 10C, 25C, 30C, 40C , 45C for 2.5 hours), pH (from 5.0 to 10.0) , salinity (the concentration of NaCl at 0.01 % , 0.05% , 0.5%, 1.0%, 1.5%, respectively) and UV (under UV for a period from 10 to 150 seconds) on the induction of lytic activity of the lysogeny phage in five strains of symbiotic Xenorhabdus and Photorhabdus bacteria were examined. The solid monoxenic culture system of the corresponding host entomopathogenic nematodes ( Steinernema carpocapsae A24, S. car-pocapsae ALL, S. feltiae English, S. feltiae SN, Heterorhabditis bacteriophora H06) was also examined for lysogenic phage. The incubation of the exponential growth stage of the five bacterial strains under the above conditions did not induce the lytic activity of the phage. The results showed that during mass production the conventional culture conditions hardly induce the lysogenic phage.
