摘要
目的 探讨中药紫草水煎液对成年小鼠及胚胎鼠的遗传毒性。 方法 采用小鼠骨髓嗜多染红细胞微核实验法、小鼠胚胎肝细胞转移微核实验法。 结果 成年小鼠与孕鼠分别灌胃紫草水煎液剂量为 1.0 g/ kg,诱发小鼠骨髓和胎鼠肝嗜多染红细胞的微核率分别为 ( 3.8± 1.0 )‰ ,( 3.8± 1.4 )‰ ,与阴性对照组 ( 2 .5± 1.5 )‰ ,( 2 .8± 1.1)‰比较 ,差异无显著性 ( t1 =0 .9,t2 =2 .1,P>0 .0 5 ) ,5 .0 g/ kg时就可诱发微核率阳性 ,微核率分别为 ( 4 .6± 2 .1)‰ ,5 .9± 1.7)‰ ,与阴性对照组比较 ,差异有显著性 ( t1 =2 .4 ,t2 =4 .3,P<0 .0 1) ;若灌胃剂量达 10 .0 g/ kg、 2 0 .0 g/kg时 ,微核率分别为 ( 7.1± 1.8)‰ ,( 8.8± 2 .2 )‰ ,( 9.3± 1.8)‰ ,与阴性对照组相比差异有显著性 ( t1 =5 .8,t2 =9.7,t3=8.4 ,P<0 .0 1)。 结论 一定剂量的中药紫草水煎液对小鼠遗传物质具有潜在的遗传毒性 ,并呈剂量 -反应关系。故在临床对于用药剂量及不同人群 。
Objective To detect genetic toxicity of Chinese drug Arnebia decoction in adult and fetal mice. Methods Micronucleus test was employed on adult mice of bone marrow cells and fetal mice of hepatocytes. Results The frequency of micronucleated polychromatic erythrocytes on adult and pregnant mice were (3 8±1 0) ‰ and (3 8±1 4) ‰ respectively,after gastric perfusion of Arnebia decoction in dosage of 1 0 g/kg.There was no significant difference in comparison with negative control groups (t 1=0 9,t 2=2 1,P>0 05).But significant micronucleus induction at the higher dose of 5 0 g/kg,10 0 g/kg and 20 0 g/kg were showed on adult and pregnant mice (7 1±1 8) ‰,(8 8±2 2) ‰,(9 3±1 8) ‰.Respectively compared with the negative controls (t 1=5 8,t 2=9 7,t 3=8 4,P<0 01). Conclusion A genotoxic potential of Arnebia decoction was demonstrated in our study and was dose related.It should be aware the effects in clinical practice especially among pregnant population.
