摘要
以短枝木麻黄 4个无性系幼枝茎段为外植体 ,在不同NaCl浓度的培养基上进行愈伤组织诱导 ,观察不同盐浓度对木麻黄外植体愈伤组织分化的影响。试验结果表明 :培养基中NaCl浓度达到 0 .0 6 4mol/L(0 .374 % ) ,比正常浓度高出 32 0倍的情况下木麻黄外植体表现出很强的耐盐性 ,仍然能诱导出正常的愈伤组织 ;不同的NaCl浓度的各个处理之间愈伤组织的诱导率、大小、出芽率、继代愈伤组织的大小均没有显著差异 ;但无性系之间的愈伤组织的诱导率及愈伤组织的大小均有极显著差异 ;不同继代间愈伤组织的大小有极显著差异。
The calli of four clones of Casuarina equiestifolia were induced in media of different NaCl concentration and the effects of NaCl concentration on calli were observed. The results showed that:the explants of Casuarina equisetifolia put up very strong salt resistance in tissue culture. When the NaCl concentration of media had reached 0.064 mol/L, namely the NaCl concentration of media had exceeded 320 times than normal one, calli could still be induced sucessfully; The callus inducement percentage, callus size, budding percentage and callus size of multiplication had not significant difference among different NaCl concentration treatments; The difference of callus inducement percentage,callus size were very significant in four clones;The difference of callus size was very significant in different multiplications.
