三七皂甙对兔髂动脉再狭窄模型血管直径及c-jun基因表达的影响 被引量:7

摘要 目的 探讨三七皂甙对兔髂动脉再狭窄模型的预防作用及其疗效 ,初步探讨其机制。方法 成年健康白兔 30只 ,随机分为二组 ,经球囊血管内膜剥脱法制成髂动脉再狭窄动物模型。实验组给予路路通注射液 (含三七皂甙 2 5mg/ml) 5ml,对照组给予同量生理盐水。 30d后重复造影 ,观察血管影像学改变 ;处死动物 ,取出手术段血管 ,采用RT PCR法检测血管中膜c fos、c jun基因的表达水平。结果 造影可见兔髂动脉经球囊过度扩张后 ,30d后实验组髂动脉有 30 %左右的狭窄 ,对照组有 60 %~ 70 %的狭窄 ,程度重于实验组 (P <0 0 5)。经RT PCR法检测 ,实验组c jun表达水平低于对照组 (P <0 0 5) ,而c fos表达未得到阳性结果。证明c jun表达与扩张前血管造影直径和 30d后血管造影直径之差存在相关性 (r =0 7394)。结论 三七皂甙能够在一定程度上抑制平滑肌细胞的增殖和迁移 ,从而抑制PTCA后再狭窄的发生 ,其机制可能与抑制原癌基因的表达 ,从而直接抑制平滑肌增殖的细胞周期有关。c jun的表达水平与再狭窄有关。 Objective The purpose of this study was to observe the effect of Totle Spaonins of Panax Notoginseng (PNS) on restenosis model of rabbits' iliac artery, evaluated the effect and mechanism.Methods 30 adult healthy rabbits were divided randomly into two groups.Right iliac artery of all rabbits were injured by ballon denuded-endothelizagion.Then E.group was injected Lulu Tong injection (comprise PNS 25mg/ml) 5ml, bid, iv , control group was injected equal 0.9% sodium chloride, bid, iv.After 30 days, vessel angigraphy was performed again, took photographs and recorded injured vessel diameter.Then all rabbits were executed.Take out injured vessel, it was stripped ectoblast, measured proto-oncogene c-jun, c-fos by reverse transcription-polymerase chain reaction (RT-PCR).Results The result of vessel angiography indicates: after 30 days, the target vessel of E.groups stenosis 30% in average,the control groups 60%~70% in average. Vessel average diameter of E.group was larger than control group ( P <0.05). Molecular biology result indicates: expression of c-jun was weaker in E.group than control group ( P <0.05), but the expression of c-fos was negative, and proved the correlation between expression of c-jun and restenosis.Conclusions (1) PNS can prevent the SMC's proliferation and removal, then inhibit restenosis of rabbit's iliac artery in some extent.The mechanism of it was correlated to inhibit the expression of oncogene thereby inhibit the cell period of SMC.(2)The expression of c-jun proto-oncogene is correlated to restenosis.
