摘要 为探讨PDTC对IL 1β诱导的大鼠软骨细胞生成NO的影响 ,寻找抑制NO生成的新途径。分离并培养大鼠软骨细胞 ,加入不同度的PDTC ,以IL 1β刺激软骨细胞 ,RT PCR检测iNOS的mRNA表达 ,NO检测试剂盒检测NO的含量。结果显示PDTC能显著抑制IL 1β诱导的iNOSmRNA的表达及NO的生成且呈剂量依赖性。表明PDTC能抑制IL 1β诱导的NO生成 。 The objective of the paper is to study the influence of PDTC on the production of nitric oxide (NO) by IL-1β-induced chondrocytes and to find a new way to inhibit the production of NO. Rat chondrocytes were separated, cultivated in the medium to which varied-concentrated PDTC was added and then stimulated by IL-1β; mRNA expression of iNOS was detected by RT-PCR; and NO level was detected by NO-detecting kit. The results showed that PDTC could significantly inhibit IL-1β-induced mRNA expression of iNOS and NO production and this function was dose-dependent, suggesting that PDTC functions to inhibit the NO production induced by IL-1β, which could provide the new idea to treat osteoarthropathy through inhibiting NO and protecting cartilage.
