摘要 目的 :观察抑癌基因PTEN编码产物在胃癌及癌前病变中的表达 ,探讨PTEN表达与胃癌发生的关系。方法 :应用S P免疫组化方法检测各种胃粘膜病变中PTEN编码产物表达。结果 :PTEN编码产物在正常胃粘膜、慢性浅表性胃炎、无肠化萎缩性胃炎、伴肠化萎缩性胃炎、中度异型增生、重度异型增生、早期胃癌和进展期胃癌 (各6 0例 )中的阳性表达率分别为 10 0 ,98.3,91.6 ,78.3,75 .0 ,6 3.3,6 1.7,4 3.3%。胃癌中肠型胃癌 76例 ,弥漫型胃癌4 4例 ,PTEN表达率分别为 6 0 .5 %和 38.6 %。结论 :PTEN基因编码蛋白在胃癌发生过程中进行性下调 ,PTEN蛋白表达可作为判定胃癌生物学行为的客观指标。 Objective: Our aim was to observe the expression of PTEN protein in gastric cancer and precancerous lesion, thus to investigate the relationship between PTEN expression and the pathogenesis of gastric cancer. Methods: We detected the PTEN protein expression with SP immunohistochemistry method in cases of gastric cancer and precancerous lesion. Results: The expression rate of PTEN product in normal gastric mucosa was 100%. In cases of chronic superficial gastritis, atrophic gastritis without intestinal metaplasia, atrophic gastritis with intestinal metaplasia, moderate dysplasia, severe dysplasia, gastric cancer of early and advanced stage, the expression of PTEN rate was 98.3%, 91.6%, 78.3%, 75.0%, 63.3%, 61.7%, 43.3%, respectively. Among the 120 patients with gastric cancer, 76 were intestinal type, the expression rate of PTEN protein was 60.5%, 44 were diffused type, the expression rate was 38.6%. Conclusion: The expression of PTEN protein was down regulated in the process of gastric cancer. The PTEN protein can be used as a maker to evaluate the biological behavior of gastric cancer.
