摘要
目的 通过观察氟(F^-)对牙胚细胞增殖及相关调控蛋白表达的影响,探讨氟斑牙的发生机制。方法 采用人牙胚体外器官培养模型,组织学观察低浓度(10mg/L)和高浓度(25mg/L)F^-对细胞增殖的影响,免疫组化及图像分析技术检测F对细胞周期蛋白(Cyclin)D1和增殖细胞核抗原(PCNA)表达的影响。结果 F^-处理组细胞增殖受抑,Cyclin D1和PCNA的表达显著降低(P<0.01)。结论 F^-可能通过抑制Cyclin D1和PCNA的表达引起细胞增殖的抑制,导致牙胚发育异常。
Objective To investigate the effects of fluoride on human tooth-germ fell proliferation and expression of related regulators for further researches on the mechanisms of dental fluorosis. Methods By using an organ culture model of human tooth-germ in vitro, the effects of low( 10 mg/L) and high(25 mg/L) concentrations of fluorine on cell proliferation were observed in histology and the expression of Cyclin D1 and proliferating cell nuclear antigen (PCNA) were detected by immunohistochemistry staining and image analysis technique. Results Fluoride inhibited cell proliferation and evidently decreased the expression of Cyclin D1 and PCNA. Conclusion Fluoride may inhibit tooth-germ cell proliferation by restraining the expression of Cyclin D1 and PCNA, leading to the abnormal tooth development.
