摘要
利用反相悬浮聚合技术合成了珠状甲基丙烯酸缩水甘油酯 N 乙烯吡咯烷酮 N,N′ 亚甲基双丙烯酰(GMA NVP MBAA)三元GNM共聚物.研究表明,GMA与NVP质量比为1∶10,MBAA为单体总量50%合成的GNM(50)固定青霉素酰化酶,固定化酶水解青霉素G钾盐活性达491U/g.固定化酶经4次使用后活性达到稳定值为444U/g,再经14次使用,活性几乎不变.同时,考察了水解反应温度、pH值对固定化酶活性的影响.
The beaded glycidyl methacrylate (GMA)Nvinyl2pyrrolidone (NVP)N, N′methylenebis(acrylamide) (MBAA) copolymers (GNM) were synthesized by inverse suspension polymerization and used as support for the immobilization of penicillin G acylase. The activity of enzyme immobilized on GNM was about 491 U/g, when the polymer was synthesized at w (NVP)/w (GMA) = 1/10 with 50% crosslinking agent. After it was used to hydrolyze penicillin G for 4 times, the activity of the immobilized enzyme reached relatively steady value (444 U/g) from 491 U/g, and was hardly changed in the following repeatedly use for 14 times.
